Abstract:
Objective:To explore the relationship between the expression levels of serum microRNA-491-3p (miR-491-3p) and miR-412-3p and the prognosis of colorectal cancer (CRC) patients and their predictive value.Methods:A total of 110 CRC patients (CRC group) and 98 healthy subjects (control group) were collected as observation objects.The expression levels of serum miR-491-3p and miR-412-3p were detected by qRT-PCR.Aaccording to the follow-up results,CRC patients were divided into survival group (73 cases) and death group (37 cases).Clinical data and serum expression levels of miR-491-3p and miR-412-3p were compared.ROC curve was applied to analyze the predictive value of serum miR-491-3p and miR-412-3p levels in mortality of CRC patients,multivariate Cox regression was applied to analyze factors that affected the prognosis of CRC patients.Results:Compared with the control group,the miR-491-3p expression level in the CRC group was decreased,while the miR-412-3p expression level was increased (P<0.05).Compared with patients without lymph node metastasis and TNM stage I-II,patients with lymph node metastasis and TNM stage III-IV hada lower miR-491-3p level and a higher miR-412-3p level (P<0.05).Compared to the survival group,the proportion of patients with lymph node metastasis and TNM stage III-IV in the death group increased,and the expression level of miR-491-3p was decreased,while the expression level of miR-412-3p was increased (P<0.05).The AUC of miR-491-3p and miR-412-3p levels,and combined in predicting 3-year mortality in CRC patients was 0.842,0.917,and 0.967,respectively.miR-412-3p was an independent risk factor for 3-year mortality in CRC patients,while miR-491-3p was a protective factor (P<0.05).Conclusion:The expression level of miR-491-3p in serum of CRC patients decreases,while the expression level of miR-412-3p increases,they are related to patients'prognosis,and both have certain predictive value for the 3-year prognosis of CRC patients.
