Abstract:
Objective:To detect the proliferation,apoptosis and migration of tumor cells and the expression of Wnt/β-catenin and PI3K-AKT signaling pathways proteins of ECA109 cells by using exosomes derived from esophageal cancer,and to explore the role of exosomes in the development of esophageal cancer cells.Methods:Exosomes were extracted from peripheral blood serum of patients with esophageal cancer by ultra-high speed centrifugation method and co-cultured with ECA109 cells to detect the changes of cell proliferation,apoptosis,migration and other biological behaviors.The expressions of AKT and β-catenin were detected by Western-blot.Results:Transmission electron microscopy,particle size measurement and marker protein results confirmed that the extracted serum exosomes from patients with esophageal cancer met the identification criteria.Compared with the blank cell group,after the intervention of exosomes,the proliferation,migration and G2+S phase of esophageal cancer cells were significantly increased,and the apoptosis rate was significantly decreased.The phosphorylation level of AKT protein in the exosome group was significantly higher than that in the blank cell group,and the expression of β-catenin was significantly lower.Conclusion:Exosomes derived from esophageal cancer can promote the activation of AKT,promote the proliferation and migration of tumor cells,and inhibit cell apoptosis,suggesting that the role of tumor-derived exosomes in tumor development should not be ignored.
