Abstract:
Objective:To investigate the expression of phospholipase PLCD3 in colorectal cancer tissues and cells,its impact on colorectal cancer cell proliferation,invasion,migration,and the underlying mechanisms.Methods:Immunohistochemistry was employed to analyze PLCD3 expression in colorectal tissues,and prognosis data were obtained from Kaplan-Meier Plotter databases.The expression levels of PLCD3 in human immortalized colonic epithelial cells (NCM460) and colorectal cancer cells were confirmed via RT-qPCR,and genetic tools were utilized to interfere with SW620 and SW480 cells.Proliferation,migration,and invasion were assessed using colony formation assay,CCK8 proliferation assay,Scratch assay,and Transwell assay.Changes in EMT-related proteins were detected by Western blot.Results:PLCD3 expression was significantly higher in colorectal cancer tissues compared to adjacent tissues(P<0.05),and high PLCD3 expression was strongly correlated with poor prognostic survival(P<0.001).Knockdown of PLCD3 inhibited SW620 cell proliferation,invasion,and migration,and the relative expression of N-cadherin,MMP2,and MMP9 was significantly reduced (all P<0.01),and the expression level of E-cadherin was obviously increased (P<0.001).Overexpression of PLCD3 promoted the proliferation,invasion and migration of SW480 cells,and the relative expression of N-cadherin,MMP2 and MMP9 was greatly elevated (all P<0.001),while the expression level of E-cadherin was significantly reduced (P<0.000 1).Conclusion:Colorectal cancer exhibits up-regulated PLCD3 expression,which may promote colorectal cancer cell proliferation,invasion,and migration by regulating the EMT process.
