Abstract:
Objective:To analyze the relationship between the expression level of long non-coding RNA(LINC00894) in lung adenocarcinoma and the clinicopathological features of patients,and to investigate the effect of overexpression of LINC00894 on the biological capabilities of lung adenocarcinoma cells.Methods:Using the GEPIA2 database,we examined the expression of LINC00894 in lung adenocarcinoma tissues and its relationship with patient prognosis,and the association between LINC00894 and the clinical pathological characteristics of patients was analyzed using the TCGA database.The aim was to utilize real-time fluorescence quantitative PCR (RT-qPCR) to analyze the expression of LINC00894 in both cancerous and adjacent tissues by collecting tissue samples from 20 lung adenocarcinoma patients.The RT-qPCR method was used to detect the presence of LINC00894 in both the human normal bronchial epithelial cell line HBE,as well as the lung adenocarcinoma cell line H1299.H1299 cells were cultured in vitro and divided into Vector group transfected with empty vector pEGFP-C1 and LINC00894 group transfected with overexpressed vector PEGFP-C1-LINC00894.The CCK-8 assay,scratch assay,Transwell assay,and flow cytometry were utilized to measure cell proliferation,migration,invasion,and apoptosis.Results:LINC00894 was low expressed in lung adenocarcinoma tissues and cells and was significantly associated with tumor T stage and primary treatment outcomes[disease progression(PD),disease stabilization(SD),partial response(PR),complete response(CR)].The expression of LINC00894 decreases as both the later T stage and the worse treatment outcome occur in lung adenocarcinoma.After artificially boosting the levels of LINC00894 in vitro,there was a notable reduction in the proliferation,migration,and invasion abilities of the cells,along with an increase in apoptosis.Conclusion:The occurrence and development of lung adenocarcinoma may be strongly influenced by LINC00894.Elevated expression of LINC00894 in lung adenocarcinoma cells inhibits their growth,invasion,and migration,while also inducing apoptosis.
