Abstract:
Objective:To investigate the effects of lncRNA HOT transcript antisense RNA (HOT transcript antisense RNA,HOTAIR) on the proliferation,migration and invasion of non-small cell lung cancer (NSCLC) and the related molecular mechanisms.Methods:The expression levels of lncRNA HOTAIR and miR-1 in adjacent tissues,lung cancer tissues and lung cancer cells A549 and NCI-H596 were detected by RT-qPCR.After over-expression and knockdown of lncRNA HOTAIR,CCK-8 assay,Transwell and cell scratch assay were used to detect the proliferation,migration and invasion of non-small cell lung cancer cells.The interaction between HOTAIR and miR-1 was verified by RNA binding protein immunoprecipitation assay (RIP).Subsequently,rescue experiments were performed to detect the effects of knockdown of miR-1 on the proliferation,migration and invasion of A549 cells.Results:Compared with adjacent tissues,the expression of lncRNA HOTAIR in lung cancer tissues was increased,and the expression of miR-1 was decreased (P＜0.05).In non-small cell lung cancer cell lines,lncRNA HOTAIR and miR-1 also showed the same expression trend.Knockdown or over-expression of lncRNA HOTAIR inhibited or promoted the proliferation,migration and invasion of A549 cells (P＜0.05).lncRNA HOTAIR can target miR-1,and interfering with miR-1 can partially reverse the inhibitory effect of silencing lncRNA HOTAIR on the proliferation,migration and invasion of A549 cells.Conclusion:The results of this study indicate that lncRNA HOTAIR can promote the proliferation,migration and invasion of NSCLC A549 cells by targeting miR-1,which is a potential molecular target of NSCLC.
