Abstract:
Objective:To investigate the effects of A disintegrin and metalloproteinase 8(ADAM8) on the proliferation and migration of colon cancer cells,as well as to elucidate its underlying mechanisms.Methods:Expression levels of ADAM8 in colon cancer tissues and normal tissues were analyzed using the cancer genome atlas(TCGA) database.The relative expression of ADAM8 in colon cancer cell lines(CaCO-2,HCT8,HT29,HCT116,SW620,LOVO) was determined via quantitative fluorescence PCR.Colon cancer cell lines overexpressing ADAM8 were established using lentiviral transduction.Real-time quantitative polymerase chain reaction(RT-qPCR) and Western blot were employed to verify ADAM8 expression levels.The in vitro proliferative capacity of colon cancer cells overexpressing ADAM8 was assessed using CCK-8 assays and colony formation assays.The effects of ADAM8 overexpression on in vitro migration of colon cancer cells were evaluated using Transwell and wound-healing assays.Western blot was utilized to determine the relative expression levels of EMT-associated proteins (Snail,Vimentin,N-cadherin,and E-cadherin) and proteins related to the signal transducer and activator of transcription 3 (STAT3) signaling pathway (STAT3 and p-STAT3).Results:Compared with normal tissues,ADAM8 expression was elevated in colon cancer tissues (P<0.001).The relative expression of ADAM8 was lower in HCT116 and LOVO colon cancer cell lines (P<0.05).The results from CCK-8 and colony formation assays indicated that overexpression of ADAM8 promoted the proliferative capacity of colon cancer cells (P<0.05).Transwell and wound-healing assay results demonstrated enhanced migration of colon cancer cells with ADAM8 overexpression (P<0.001).Western blot findings revealed that overexpression of ADAM8 downregulated E-cadherin,upregulated Snail,N-cadherin,and Vimentin,and activated the STAT3 signaling pathway (P<0.05).Conclusion:ADAM8 can promote the proliferation and migration of colon cancer cells through the activation of the STAT3 signaling pathway,suggesting ADAM8 may be a potential therapeutic target for colon cancer treatment.
