Abstract:
Objective:To investigate the expression of cyclin B2 (CCNB2),SRY-box transcription factor 11 (Sox11) and miR-21 in endometrial cancer and clinical significance.Methods:Patients with endometrial cancer (EMC group) and endometrial benign disease (CON group) who were diagnosed and treated from January 2015 to December 2017 were selected as research objects,and real-time fluorescent quantitative PCR (RT-qPCR) was used to detect CCNB2,Sox11 and miR-21 expression.Western blot was used to detect the protein expression of CCNB2 and Sox11.The differences in the expression of CCNB2,Sox11 and miR-21 in different clinicopathological factors were compared.The ROC curve was used to analyze the efficacy of serum CCNB2,Sox11 and miR-21 in predicting the death of patients with endometrial cancer.Logistic regression analysis evaluated the risk factors for poor prognosis in endometrial cancer.Kaplan-Meier method was used to analyze the relationship between the expression of CCNB2,Sox11 and miR-21 and the survival time.Results:The relative expressions of CCNB2,Sox11 and miR-21 in tumor tissues of EMC group were significantly higher than those in paracancerous tissues and CON group (all P<0.05).The relative expressions of CCNB2,Sox11 and miR-21 in Ishikawa,ECC-1,Hec50co and RL95-2 endometrial cancer cells were significantly higher than those in normal human endometrial epithelial cells HEMEC (all P<0.05).There were significant differences in the expression of CCNB2,Sox11 and miR-21 in the tumor tissue of EMC group with the degree of differentiation,histological grade,depth of myometrial invasion,lymph node metastasis and FIGO stage (all P<0.05).The cut-off values of CCNB2,Sox11 and miR-21 in predicting the death of endometrial cancer patients were 1.2,0.8,1.5.The sensitivities were 0.771,0.827,0.833,and the specificities were 0.785,0.762,0.701,respectively.Poorly differentiated,histological grade G3,depth of myometrial invasion>1/2,lymph node metastasis,stage III,relative expression of CCNB2≥1.2,relative expression of Sox11≥0.8,relative expression of miR-21≥1.5 were the risk factors for death of endometrial cancer patients (P<0.05).The median survival time of endometrial cancer patients with CCNB2 expression≥1.2,Sox11 expression≥0.8 and miR-21 expression≥1.5 was significantly lower than that of patients with CCNB2 expression<1.2 or Sox11 expression<0.8 or miR-21 expression<1.5[median survival (27.2±6.4) months vs (32.5±6.8) months,Logrank=9.821,P<0.05].Conclusion:The expressions of CCNB2,Sox11 and miR-21 are significantly increased in patients with endometrial cancer,which are closely related to the degree of differentiation,histological grade,depth of myometrial invasion,lymph node metastasis,FIGO stage,survival period and risk of death,which can be used as the markers of endometrial cancer condition and prognosis assessment.
