Abstract:
Objective:To explore the role of ERK-VEGF/MMP-9 signalling pathway in regulating the adverse biological behaviours of hepatocellular carcinoma HepG2 cells.Methods:Human hepatocellular carcinoma HepG2 cells were cultured in vitro,and 54 dishes were divided into control,blocking and transfection groups,each with 18.No intervention was performed in the control group.The blocking group was treated with a specific ERK-VEGF/MMP-9 signalling pathway inhibitor,GDC-0994 and the transfection group was transfected with ERK1 plasmids.The proliferation of HepG2 cells was detected by MTT.Cell cycle and apoptosis were detected by flow cytometry.Transwell assay to analyse HepG2 cell migration and invasion.Western blot to detect ERK,VEGF and MMP-9 protein expression.Results:Compared with the control group,the blocking group showed a decrease in cell proliferation rate,an increase in the percentage of cells in S phase and G2/M phase,an increase in apoptosis rate,and a decrease in the number of cells migrating and invading (P<0.05),while the transfection group showed an increase in cell proliferation rate,a decrease in the percentage of cells in S phase and G2/M phase,a decrease in apoptosis rate,and an increase in the number of cells migrating and invading (P<0.05).Compared with the control group,ERK,VEGF and MMP-9 protein expression decreased in the blocking group (P<0.05).ERK,VEGF and MMP-9 protein expression increased in the transfection group (P<0.05).Conclusion:Blockade of ERK-VEGF/MMP-9 signalling pathway inhibits proliferation,migration and invasion and promotes apoptosis in HepG2 cells.
