Abstract:
Objective:To investigate the expression of homeobox B9(HOXB9) and cell division cycle-associated 5(CDCA5) in uterine corpus endometrial cancer(UCEC) tissue,and analyze their clinical application value as a poor prognosis warning model for UCEC.Methods:The expression of HOXB9 and CDCA5 in 174 cases of UCEC tissues and 91 cases of normal control tissues was analyzed by Gene Expression Profiling Interactive Analysis(GEPIA) online website.Using the Kaplan-Meier Plotter online website to analyze the expression of HOXB9(172 patients with UCEC) and CDCA5(542 patients with UCEC) and their relationship with survival in UCEC patients.Furthermore,from February 2017 to December 2019,fifty-six cases of UCEC admitted to our hospital were selected as the study subjects.Detect the levels of HOXB9 and CDCA5 in tissues and analyze their relationship with the clinical and pathological features of UCEC.Using a multivariate logistic regression model to analyze the risk factors for poor prognosis in UCEC,and a risk warning model was constructed and transformed into a risk scoring system.The receiver operating characteristic(ROC) curve was used to evaluate the effectiveness in predicting poor prognosis of UCEC.Results:According to data from the GEPIA online website,the expression levels of HOXB9 and CDCA5 proteins in UCEC tissue were higher than those in the normal control group,with statistical significance(P<0.05).Survival curve analysis showed that an increase in HOXB9 and CDCA5 expression levels resulted in a decrease in patient survival rate(Logrank P of 0.003 4 and 0.000 27,respectively).The expression levels of HOXB9 and CDCA5 in the cancer tissues of 56 UCEC patients were significantly higher than those in adjacent normal tissues,with statistical significance(P<0.01).The high expression of HOXB9 and CDCA5 were statistically significant among patients with different FIGO stages,tumor differentiation,invasion depth,and lymph node metastasis(χ2=5.364,6.103,8.812,7.654;χ2=6.222,8.319,7.754,7.108,all P<0.05).Regression analysis showed that FIGO stage Ⅲ-Ⅳ,poor tumor differentiation and high expression of HOXB9 and CDCA5 were risk factors for poor prognosis of UCEC(all P<0.05).The area under the curve of the warning model was 0.818.The sensitivity was 0.809 and specificity was 0.829.Conclusion:High expression of HOXB9 and CDCA5,FIGO stage Ⅲ-Ⅳ and poor tumor differentiation are closely related to poor prognosis of UCEC tissue.The constructed warning model has important reference value for predicting poor prognosis of UCEC patients.
