Abstract:
Objective:To investigate the effects of MMI-166 on the migration and invasion of human pancreatic cancer cell lines PANC-1 and SW1990.Methods:PANC-1,SW1990 cells were cultured in vitro,which were treatment with different concentrations of MMI-166.Cell migration and invasion were detected by scratch assay and Transwell assay.The expressions of proteins of MMP-2 and MMP-9 were compared in different groups of PANC-1 and SW1990 cells.Results:Compared with the control group,the scratch assay and Transwell assay showed that MMI-166 decreased cell migration and invasion with increasing concentration,with statistical difference (P<0.05).Western blot and immunofluorescence showed MMI-166 significantly inhibited the expression of MMP-2 and MMP-9 protein in a dose-dependent manner,with statistical difference (P<0.05).Conclusion:MMI-166 inhibited the MMP-2 and MMP-9 activity in pancreatic cancer cells and it is useful for the regulation of pancreatic cancer cells migration and invasion.
