Abstract:
Objective:To investigate the effect of Trim37 through Wnt/β-catenin pathway on the development of breast cancer and its mechanism.Methods:The expression of Trim37 in breast cancer tissues,adjacent normal tissues,MCF-7 cells and MCF-10A cells were detected by RT-qPCR and Western blot.MCF-7 cells were transfected with pcDNA-Trim37 or Trim37 siRNA,the effects of overexpression and downregulation of Trim37 on cell proliferation were detected by CCK-8 assay,the effects of overexpression and downregulation of Trim37 on cell invasion were detected by Transwell assay,and the effects of overexpression and downregulation of Trim37 on cell migration were detected by scratch healing assay.Western blot was used to detect the effects of overexpression and downregulation of Trim37 on the expression of EMT-related proteins (E-cadherin,N-cadherin and Snail),β-catenin,CyclinD1,p-ERK,p-AKT and p-FAK in cells.Wnt/β-catenin pathway inhibitor (XAV939) acted on MCF-7 cells,the proliferation,invasion,migration,and EMT of MCF-7 cells were detected.Results:The expression of Trim37 mRNA and protein in breast cancer tissues was significantly higher than that in paracancer tissues (P<0.01).Compared with MCF-10A cells,the Trim37 mRNA and protein expression levels in MCF-7 cells were significantly increased (P<0.01).The overexpression of Trim37 promoted the proliferation,invasion,migration and EMT of MCF-7 cells,the protein expression levels of β-catenin and CyclinD1 increased significantly.While the downregulation of Trim37 inhibited the proliferation,invasion,migration and EMT of MCF-7 cells,the protein expression levels of β-catenin and CyclinD1 decreased significantly,while the overexpression and downregulation of Trim37 had no effect on the phosphorylation levels of ERK,AKT and FAK proteins.XAV939 inhibited the proliferation,invasion,migration and EMT of MCF-7 cells after overexpressing Trim37.Conclusion:Overexpression of Trim37 promotes the development of breast cancer by activating the Wnt/β-catenin pathway to induce EMT.
