Abstract:
Objective:To investigate the exact role and molecular mechanism of lncRNA NEAT1 in the progression of ovarian cancer,and to provide a new inhibition for the screening of biomarkers and targeted therapy of ovarian cancer.Methods:Real-time fluorescent quantitative PCR (qRT-PCR) was used to detect the expression levels of miR-335-5p and BCL2L2 in ovarian cancer cells and tissues.MTT and Transwell assays were performed to detect cell proliferation,migration and invasion abilities.Western blot was used to analyze the expression level of BCL2L2.The dual luciferase reporter assay confirmed the interaction between miR-335-5p and NEAT1/BCL2L2.Results:The results showed that the expression of BCL2L2 was increased,while the expression of miR-335-5p was decreased in ovarian cancer tissues compared with adjacent tissues,and the differences were statistically significant (P<0.05).Overexpression of miR-335-5p inhibited the expression of BCL2L2 in ovarian cancer cells,while inhibition of miR-335-5p increased the expression of BCL2L2 in ovarian cells (P<0.05).Furthermore,it was found that miR-335-5p could inhibit the proliferation and migration of ovarian cancer cells,and the difference was statistically significant (P<0.05).In addition,lncRNA NEAT1 was also highly expressed in ovarian cancer tissues and promoted proliferation,migration and invasion of ovarian cancer cells,and the difference was statistically significant (P<0.05).Conclusion:lncRNA NEAT1 can regulate the proliferation and migration of ovarian cancer cells through miR-335-5p/BCL2L2 pathway,which may be a potential target for early screening or targeted therapy of ovarian cancer.
