Abstract:
Objective:To investigate the effect of sophoridine on apoptosis of prostate cancer PC3 cells and the possible mechanism of promoting apoptosis.Methods:The inhibitory effect of sophoridine on the proliferation of PC3 cells was analyzed by MTT method.Trypan blue staining assay was used to detect the effect of sophoridine on the growth and proliferation of PC3 cells.Hoechst 33342 staining and flow cytometry were used to detect the effect of sophoridine on apoptosis of PC3 cells.Western blot detected the effect of sophoridine on the expression of Caspase-3,Bax,Bcl-2,and apoptosis signaling pathway proteins p-JNK,p-p38,p-ERK in PC3 cells.Results:With the increase of sophoridine concentration,the inhibition rate of PC3 cells was gradually increased in a dose-dependent manner,and the proliferation of PC3 cells was significantly inhibited(P<0.05).The apoptosis rate of sophoridine group was significantly increased,and the expressions of Caspase-3 and Bax apoptosis-promoting proteins were significantly increased,while the expressions of Bcl-2 anti-apoptosis-inhibiting proteins were significantly decreased(P<0.05).The expression of MAPK signaling pathway protein p-JNK was significantly increased,while the expression of p-p38 and p-ERK had no significant differences(P>0.05).JNK inhibitor SP600125 reversed the promoting effect of sophoridine on apoptosis of PC3 cells(P<0.05).Sophoridine could reduce tumor volume and mass in nude mice(P<0.05).Conclusion:Sophoridine can inhibit the proliferation and promote the apoptosis of PC3 cells in prostate cancer,and has good antitumor activity.It may mediate apoptosis of prostate cancer PC3 cells through JNK/MAPK signaling pathway.
