Abstract:
Objective:To explore the differential expression profile of serum exosome microRNA (miRNA) in patients with papillary thyroid carcinoma (PTC) lymph node metastasis (LNM) under high iodine and normal iodine levels,and to provide reference for the early differential diagnosis of PTC LNM at different iodine levels.Methods:Five patients in the high iodine and normal iodine PTC LNM+ group and PTC LNM- group were selected for high-throughput sequencing of serum exosome miRNA.The common differential expression of exosome miRNA in PTC LNM+ group at different iodine levels was screened.The target gene prediction,GO and KEGG enrichment analysis were carried out.Results:There were five kinds of differential expression of exosome miRNAs in the high iodine and normal iodine PTC LNM+ groups.The expression of hsa-miR-7113-5p,hsa-miR-873-3p and hsa-miR-548q was opposite at different iodine levels,and the expression of hsa-miR-616-3p and hsa-miR-5189-5p was up-regulated.The differentially expressed exosome miRNA regulated HIF-1 signaling pathway,neuroactive ligand-receptor interaction and GnRH signaling pathway.Conclusion:When serum exosome hsa-miR-7113-5p,hsa-miR-873-3p and hsa-miR-548q are used to screen PTC LNM,high iodine and normal iodine levels should be distinguished.The hsa-miR-616-3p and hsa-miR-5189-5p can be used for the early diagnosis of PTC LNM in the general population.
