Abstract:
Objective:To screen out miRNAs significantly associated with the prognosis of oral squamous cell carcinoma (OSCC) using bioinformatics analysis,and validate it through in vitro experiments.Methods:OSCC data were downloaded from The Cancer Genome Atlas (TCGA).Differentially expressed miRNAs were analyzed by R.The risk model was established using regression analysis.The prognosis of OSCC patients was predicted using Kaplan-Meier survival analysis.The reliability of the risk model was verified by receiver operating characteristic (ROC) curve and regression analysis.Biological functions of prognostic miRNAs were predicted by GO and KEGG analyses.In vitro validation of bioinformatics results was conducted through qRT-PCR and Western blot.Results:Twenty differentially expressed miRNAs in OSCC were screened out.miR-204-5p and miR-503-3p were significantly correlated with the prognosis of OSCC.The risk model constructed in this study could reliably predict the prognosis of OSCC.GO analysis showed miR-204-5p was related to cell apoptosis,proliferation and migration,while miR-503-3p was related to cell differentiation,cell cycle and GTP binding.KEGG analysis showed miR-204-5p was related to PI3K/Akt/mTOR pathway and autophagy pathway,while miR-503-3p was related to Ras pathway and HIF-1 pathway.qRT-PCR showed that the expression of miR-204-5p in OSCC cells was decreased (P<0.05),while miR-204-5p level in OSCC cells was artificially increased after transient transfection of miR-204-5p mimic (P<0.05).In addition,Western blot showed that the expression of PIK3CB in OSCC cells was decreased after miR-204-5p elevation (P<0.05).Conclusion:The results showed that miR-204-5p and miR-503-3p were effective prognostic biomarkers in OSCC.The risk model constructed by miR-204-5p and miR-503-3p could effectively predict the prognosis of OSCC patients.The expression of miR-204-5p in OSCC cells was low.After elevation of miR-204-5p,the expression of PIK3CB in OSCC cells was decreased.
