Abstract:
Objective:To analyze the lncRNA TUG1 correlation with clinicopathological characteristics,and to explore the effects of lncRNA TUG1 on the proliferation and invasion of colorectal cancer cells.Methods:Real-time quantitative PCR(RT-qPCR) was used to detect the expression level of TUG1 in colorectal cancer.Chi-square test was used to analyze the correlation between TUG1 expression level and clinical data.Kaplan-Meier survival curve and log-rank test were used to analyze the correlation between TUG1 expression level and prognosis of colorectal cancer.CCK8 experiment and Transwell experiment were used to detect the changes of proliferation or invasion ability of colorectal cancer cells after overexpression and knockdown of TUG1.Results:The expression level of TUG1 in colorectal cancer was significantly increased(P<0.05). The expression level of TUG1 is closely related to tumor size,CEA level,lymph node metastasis,distant metastasis and TNM staging(P<0.05).Kaplan-Meier survival curve and log-rank test indicated that the higher the expression level of TUG1, the worse the prognosis of patients with colorectal cancer(P<0.05).The results of CCK8 and Transwell experiments showed that compared with the control group,the expression level of TUG1 in colorectal cancer cells in pcDNA3.1-TUG1 group increased significantly(P<0.05),and the cell proliferation activity and invasion number increased(P<0.05).The expression level of TUG1 in colorectal cancer cells in siRNA-TUG1 group decreased significantly(P<0.05),and the cell proliferation activity and invasion number decreased(P<0.05).Conclusion:TUG1 is highly expressed in colorectal cancer tissues,and the higher the expression level of TUG1,the worse the prognosis of patients.Furthermore,lncRNA TUG1 promotes the proliferation and invasion of colorectal cancer cells.
