Abstract:
Objective:To investigate the effect of long non-coding RNA (lncRNA) lnc AL137789.1 on the proliferation and migration of colorectal cancer cells.Method:A total of 40 patients with colorectal cancer who were admitted to Hainan Cancer Hospital from June 2017 to January 2020 and had complete clinical data were enrolled.Surgically resected cancer tissues (colorectal cancer group) and adjacent tissues>2 cm away from cancer tissues (normal group) were collected.Quantitative real-time PCR and Western blot were used to detect the expression levels of lnc AL137789.1 and JAK2 in colorectal cancer tissues and cells.The positive rate of JAK2 in colorectal cancer tissues and adjacent normal tissues was detected by immunohistochemistry (IHC).The proliferation and migration of colorectal cancer cells were detected by EdU and Transwell assay.The correlation between lnc AL137789.1 and JAK2 was analyzed by Pearson correlation coefficient.Results:The relative expression of lnc AL137789.1 in cells and tissues of colorectal cancer group was significantly higher than that in adjacent normal group (P<0.05),and the relative expression of JAK2 in cells and tissues of colorectal cancer group was significantly higher than that in normal group (P<0.05).Compared with SH-lnc AL137789.1-NC group,SH-lnc AL137789.1 group significantly decreased the expression of JAK2 protein (P<0.05).Compared with LV-lnc AL137789.1-NC group,LV-lnc AL137789.1 group significantly promoted the expression of JAK2 protein (P<0.05).Pearson correlation coefficient analysis showed that JAK2 was positively correlated with lnc AL137789.1 group (R2=0.401 5,P<0.000 1).Compared with SH-lnc AL137789.1-NC group,SH-lnc AL137789.1 group significantly inhibited the proliferation and migration of colorectal cancer cells (P<0.05),while SH-LNC AL137789.1+JAK2 group significantly promoted the proliferation and migration of colorectal cancer cells (P<0.05).Compared with LV-lnc AL137789.1-NC group,LV-lnc AL137789.1 group significantly promoted the proliferation and migration of colorectal cancer cells (P<0.05),while LV-lnc AL137789.1+si-JAK2 group significantly inhibited the proliferation and migration of colorectal cancer cells (P<0.05).Conclusion:Lnc AL137789.1 regulates the proliferation and migration of colorectal cancer cells by structurally modifying JAK2 protein.
