Abstract:
Objective:To investigate the effect of lycorine on the proliferation of esophageal cancer cells and analyze its mechanism.Methods:The inhibition rate of cell proliferation was analyzed by cell counting kit-8 (CCK-8).Autophagy vesicles were detected by MDC staining.The levels of LC3II/LC3I,Beclin-1,transforming growth factor-β (TGF-β) and p-Smad2/Smad2 were analyzed by Western blot.The effect of 3-MA on autophagy and apoptosis regulated by lycorine was observed.The tumorigenesis experiment of nude mice verified the effect of allicin on tumor weight,tumor volume,autophagy and p-Smad2/Smad2 level in nude mice.Results:Lycorine at 2、4、8、16、32、64 μmol/L inhibited the proliferation of ECA109 cells (F= 22.412,P<0.05),and the IC50 value was (15.56±2.16)μmol/L.4,8 and 16 μmol/L lycorine promoted the production of autophagy vesicles,increased the levels of LC3II/LC3I and Beclin-1,and decreased the levels of TGF-β1 and p-Smad2/Smad2.3-MA could reverse the effects of lycorine on autophagy,apoptpsis,TGF-β/Smad signal pathway and Bcl-2/Bax signal pathway.100 mg/kg lycorine could reduce tumor volume,tumor mass,down-regulate tumor p-Smad2/Smad2 level and up-regulate LC3II/LC3I level.3-MA could reverse the effect of lycorine on tumor volume,tumor mass and the level of LC3II/LC3I and p-Smad2/Smad2.Conclusion:Lycorine can inhibit the proliferation of ECA109 cells,promotes apoptosis and has obvious anti-tumor effect on ECA109 fine cell transplanted tumor in vivo.The mechanism is related to the activation of autophagy by regulating TGF-β/Smad signal pathway.
