Abstract:
Objective:To investigate the effect of miR-497-5p on the migration and invasion of esophageal squamous cell carcinoma (ESCC) by high mobility group protein A2 (HMGA2).Methods:UALCAN and GEPIA databases were used to analyze the expression of miR-497-5p and HMGA2 mRNA in esophageal carcinoma (ESCA) and its control samples.The correlation between miR-497-5p and HMGA2 expression in ESCA samples was analyzed by starBase,a bioinformatics website,and the binding sites between them were predicted.The regulation effect of miR-497-5p on HMGA2 was confirmed by dual luciferase reporter assay.The effect of miR-497-5p on the migration and invasion of ESCC cells through HMGA2 was analyzed by Wound healing assay and Transwell assay.Results:The expression of miR-497-5p in ESCA was significantly lower than that in the control samples (P<0.05),and the expression of HMGA2 mRNA in ESCA was significantly higher than that in the control samples (P<0.05).miR-497-5p was negatively correlated with HMGA2 in ESCA samples.There were binding sites between miR-497-5p and HMGA2,and miR-497-5p directly regulated the target gene HMGA2.The migration and invasion ability of miR-497-5p mimic group were significantly lower than that of negative control (NC) group (P<0.001).The migration and invasion ability of miR-497-5p mimic+HMGA2 group were significantly increased compared with mir-497-5p mimic group (P<0.01).Conclusion:miR-497-5p regulates the migration and invasion of ESCC via HMGA2.
