Abstract:
Objective:To explore the mechanism of circVAPA in reversing chemoresistance of oral cancer cells through the miR-125b/HOXA7 axis.Methods:SCC9/DDP cells were divided into SCC9/DDP group,si-NC group,si-VAPA group,miR-125b group,miR-NC group,pcDNA-VAPA group and miR-HOXA7 group.qRT-PCR was used to detect the expression of circVAPA and miR-125b in cells.MTT method,Transwell method and flow cytometry were used to detect cell drug resistance,proliferation,invasion and apoptosis.Western blot was used to detect HOXA7 protein expression in cells.Dual-luciferase reporter gene assay verified the targeting relationship of VAPA,miR-125b and HOXA7.Results:Compared with HOK,the expression of circVAPA in SCC9 and SCC9/DDP cells was significantly increased,and the expression of miR-125b was significantly decreased (P<0.05).The expression of circVAPA in SCC9/DDP cells was significantly higher than that in SCC9 cells,and the expression of miR-125b was significantly lower than that in SCC9 cells (P<0.05).Compared with the SCC9/DDP group,the IC50 concentration,cell resistance index,cell invasion ability and HOXA7 protein expression in the si-VAPA group were significantly decreased,and the apoptosis ability was significantly increased (P<0.05).Compared with the miR-NC group,the IC50 concentration,cell resistance index,cell invasion ability and HOXA7 protein expression in the miR-125b group were significantly decreased,and the apoptosis ability was significantly increased (P<0.05).The results of targeting relationship showed that the luciferase activity of the miR-125b group was significantly lower than that of the miR-NC group (P<0.05).The expression of miR-125b in the cells of the si-VAPA group was significantly higher than that of the si-NC group (P<0.05).The expression of HOXA7 in the miR-125b group was significantly lower than that in the miR-NC group (P<0.05).Compared with the miR-125b group,the IC50 concentration,cell drug resistance index,cell invasion ability and HOXA7 protein expression in the pcDNA-VAPA group and miR-HOXA7 group were significantly increased,and the apoptosis ability was significantly decreased (P<0.05).Conclusion:Low expression of circVAPA can reverse the chemotherapy resistance of oral cancer cells by regulating the miR-125b/HOXA7 axis,inhibit the proliferation,invasion and induce apoptosis of oral cancer chemotherapy-resistant cells.
