Abstract:
Objective:To explore the expression of long noncoding RNA (lncRNA) NONHSAT070341 in cervical cancer tissues and cell lines SiHa and Caski and its effect on the biological functions of cervical cancer cells such as proliferation,migration,invasion,apoptosis and cell cycle distribution.Methods:A total of 90 patients with different stages of cervical lesions who underwent surgical resection in the department of gynecology and obstetrics,the Second Clinical Medical College of Shanxi Medical University from June 2021 to June 2022 were collected,and 30 cases of normal cervical tissue undergoing total hysterectomy due to uterine fibroids at the same time were used as control.The expression of lncRNA NONHSAT070341 in cervical cancer tissues and cell lines was detected by quantitative real-time polymerase chain reaction (qRT-PCR).The lncRNA NONHSAT070341 was knocked down by transient transfection of small interfering RNA (siRNA) fragment in SiHa and Caski cells,and the transfection efficiency was verified by qRT-PCR.The cell proliferation ability was detected by CCK-8 and plate colony formation assay.The cell cycle assay was used to detect the effect on the cycle distribution of cervical cancer cells.The scratch healing assay was used to evaluate the lateral migration ability of cells,and the Transwell assay was used to determine the longitudinal migration and invasion ability of cells.Flow cytometry analyzed cell apoptosis.Results:The lncRNA NONHSAT070341 was highly expressed in cervical cancer tissues and cells.NONHSAT070341 siRNA can reduce the expression of NONHSAT070341 in SiHa and Caski cells.After knocking down NONHSAT070341,the proliferation,migration and invasion abilities of cervical cancer cells were significantly reduced,and the cells were arrested in the G1 phase,influencing the distribution of cervical cancer cell cycle,whereas the rate of apoptosis of SiHa and Caski cells were enhanced.Conclusion:NONHSAT070341 can promote the proliferation,migration and invasion of cervical cancer cells,change the biological behavior of cervical cancer and be expected to become an emerging biomarker for the diagnosis of cervical cancer.
