Abstract:
Objective:To investigate the effect of (-)gossypol on the proliferation and apoptosis in human prostate cancer PC-3 cells in vitro. Methods: The cell proliferation was evaluated by methyl thiazolyl tetrazolium (MTT)assay. Cell ultramicro-structure was studied by transmission electron microscope. The morphology of apoptotic cells was observed by TUNEL staining , cell cycle and apoptosis were detected by flow cytometry(FCM).The Bcl-2 and Bak mRNA level were measured by semiquantitative RT-PCR. Rusults: (-)Gossypol significantly inhibited the proliferation of PC-3 cells in a dose-and time-dependent manner. The PC-3 cells were arrested in G0/G1 phase and showed typical apoptosis characteristics in FCM and transmission electron microscope analysis.(-)Gossypol down-regulated Bcl-2 mRNA and up-regulated Bak mRNA level. Conclusion: (-)Gossypol can induce apoptosis of human prostate cancer PC-3 cells in vitro and the main reason may be related with the up-regulation of Bak mRNA and the down-regulation of Bcl-2 mRNA.
