Abstract:
Objective:To analyze the expression of centromere protein A (CENP-A) in cervical cancer, its correlation with clinicopathological parameters and its clinical significance. Methods:Gene Expression Profiling Interactive Analysis (GEPIA) database and Human Protein Atlas(HPA) database were used to analyze the expression of CENP-A gene and protein in cervical cancer tissues and adjacent tissues. The expression level of CENP-A protein in cervical cancer tissues and adjacent tissues was detected by immunohistochemistry, and the relationship between CENP-A protein expression and clinicopathological parameters was analyzed.Using the Linked Omics database to explore the co-expressed genes of CENP-A,and further analyze the signal regulatory pathways that CENP-A gene may participate in the development of cervical cancer. Results:The bioinformatics analysis showed that the expression level of CENP-A in cervical cancer tissues was significantly higher than that in the adjacent tissues (P<0.05),and the expression of CENP-A protein in cervical cancer tissues is higher than that in the adjacent tissues. The results of immunohistochemistry showed that the expression level of CENP-A protein in cervical cancer tissues was higher than that in the adjacent tissues (P<0.05),and the expression of CENP-A was correlated with tumor stage and degree of invasion.Through gene co-expression analysis,KIF2C,CDC20,FAM72B and other genes were positively correlated with CENP-A expression, while PARM1,ABCA9 and SYNE1 were negatively correlated with CENP-A expression (FDR<0.05).CENP-A gene is involved in cell cycle,DNA replication and other pathways.Conclusion:CENP-A is highly expressed in cervical cancer tissues, which is related to the prognosis, occurrence and development of cervical cancer.
