Abstract:
Objective:To investigate the potential role of small nucleolar RNA 73B (SNORA73B) in laryngeal squamous cell carcinoma (LSCC) by examining its high expression and clinical significance,to explore how SNORA73B may contribute to the proliferation,migration,and invasion of LSCC cells by maintaining stem cell characteristics.Methods:Quantitative PCR (qPCR) was employed to measure the expression level of SNORA73B in LSCC tissues and cells.Furthermore,in vitro cultured cells were utilized for MTS assays,scratch healing assays,and Transwell experiments to assess the impact of SNORA73B knockdown or overexpression on cell proliferation,migration,and invasion.Additionally,the role of SNORA73B in influencing the stem cell-like characteristics of LSCC cells was evaluated using a tumor cell spheroidization assay.Results:The results obtained from qPCR analysis demonstrated a substantial increase in the expression of SNORA73B in both LSCC tissues and cells compared to the corresponding adjacent cancer tissues and control group (P<0.05).This elevated expression was found to be associated with pathological grading,lymph node metastasis,and poor prognosis in patients (P<0.05).Moreover,overexpression of SNORA73B in Tu177 cells notably enhanced their proliferation,scratch healing,migration,and invasion capabilities(P<0.05).Conversely,interference with SNORA73B led to a significant reduction in these cellular functions(P<0.05).To further investigate the impact of SNORA73B on the development of LSCC stem cell-like characteristics,laryngeal cancer spheroid cells were induced(P<0.05).The induction resulted in a notable increase in the expression of tumor stem cell markers OCT4 and SOX2 proteins (P<0.05).Additionally,SNORA73B expression was significantly upregulated (P<0.05).However,interference with SNORA73B led to a significant decrease in the number of cell spheroids(P<0.05) as well as reduced mRNA and protein expressions of OCT4 and SOX2.Conclusion:The high expression of SNORA73B appears to be associated with the development and malignant progression of LSCC.It is likely that SNORA73B plays a role in mediating stem cell-like characteristics in LSCC cells,leading to increased tumor cell proliferation and migration.
