Abstract:
Objective:To explore the effect of lncRNA DRAIC on proliferation,apoptosis,and autophagy of colorectal cancer (CRC) cells and its mechanisms.Methods:GEPIA 2.0 database was used to analyze the expression and prognostic value of DRAIC in CRC.The receiver operating characteristic(ROC) curve was employed to assess the diagnostic value of DRAIC in CRC.The UALCAN online database was used to analyze the relationship between DRAIC expression and clinicopathological parameters of CRC.The DRAIC overexpression plasmid (OE-DRAIC) was transfected into HCT-116 and HT-29 cells to overexpress DRAIC.Cell proliferation ability was assessed by CCK-8 and clone formation assays,and apoptosis was detected by flow cytometry and TUNEL assays.The expression of LC3B,Cleaved caspase-3,Cleaved PARP,and AKT/mTOR pathway proteins were detected by Western blot.After treatment with OE-DRAIC plus 3-methyladenine (3-MA,an autophagy inhibitor),the proliferation and apoptosis of CRC cells,as well as the expression of LC3B,Cleaved caspase-3,and Cleaved PARP proteins,were detected using the same methods described above.Results:The expression of DRAIC was significantly downregulated in CRC tissues,with an area under the ROC curve (AUC) of 0.781.CRC patients with low DRAIC expression had poorer overall survival.Downregulation of DRAIC was associated with clinicopathological parameters such as tumor stage,histological type,and body weight.DRAIC overexpression suppressed proliferation and promoted apoptosis of CRC cells,resulting in the upregulation of Cleaved caspase-3,Cleaved PARP,and LC3B-II/LC3B-I protein levels,and the downregulation of p-AKT and p-mTOR protein levels (P<0.05).After adding 3-MA,compared with the OE-DRAIC group,the OE-DRAIC+3-MA group showed an increased proliferation ability and decreased apoptosis rate of HCT-116 and HT-29 cells (P<0.05).The expression of LC3B-II/LC3B-I,Cleaved caspase-3 and Cleaved PARP protein in HCT-116 cells were downregulated (P<0.05).Conclusion:DRAIC is downregulated in CRC tissues and is associated with a poor prognosis in CRC patients.It inhibits cell proliferation and promotes apoptosis by enhancing autophagy of CRC cells.
