Abstract:
Objective:To study the expression of tiRNA-1_33-Gly-GCC-1 in colorectal adenocarcinoma (CAC) tissues and its impact on cell proliferation.Methods:Expression profiles of tRFs and tiRNAs in 5 pairs of CAC tissues were detected and analyzed using Arraystar nrStarTM tRNA PCR Arrays,and the expression levels of tiRNA-1_33-Gly-GCC-1 in 82 pairs of CAC tissues were verified using qRT-PCR assay.RNA in situ hybridization (RISH) was used to detect the positive expression of tiRNA-1_33-Gly-GCC-1 in 82 CAC tissues.The correlation was statistically analyzed between its positive expression and the clinical pathological characteristics and prognosis of patients.CCK-8 experiment was used to detect the effect of tiRNA-1_33-Gly-GCC-1 on the proliferation activity of colorectal cancer cells.Bioinformatics and double luciferase report experiment were used to verify how tiRNA-1_33-Gly-GCC-1 regulates TP53I11.Rescue experiment to verify the effect of the tiRNA-1_33-Gly-GCC-1/TP53I11 signal axis on the proliferation of colorectal cancer cells.The subcutaneous transplanted tumors experiment in nude mice was used to verify the effect of tiRNA-1_33-Gly-GCC-1 on the growth and proliferation of transplanted tumors.Results:The expression of tiRNA-1_33-Gly-GCC-1 was increased and mainly positive in CAC tissues,with a significantly higher number of positive cases in 54/82 (65.85%) compared to 11/82 (13.41%) in adjacent normal tissues(χ2=5.475,P=0.019<0.05).Positive expression of tiRNA-1_33-Gly-GCC-1 and tumor diameter in CAC patients(χ2=11.699),degree of differentiation(χ2=13.081) and patient survival prognosis(χ2=15.621) is closely related (all P<0.05).The results of the CCK-8 experiment showed tiRNA-1_33-Gly-GCC-1 increased the proliferative activity of CAC cells(P<0.05).Bioinformatics predicts that TP53I11 is one of the downstream target genes of tiRNA-1_33-Gly-GCC-1.The double luciferase reporter experiment suggested that tiRNA-1_33-Gly-GCC-1 could binds to TP53I11 3' UTR to inhibit its protein synthesis.The Rescue experiment result further validated the inhibitory effect of tiRNA-1_33-Gly-GCC-1 on the expression of TP53I11 mRNA and protein and its promoting effect on the proliferation of colorectal cancer cells.The subcutaneous tumor transplantation experiment in nude mice also showed that tiRNA-1_33-Gly-GCC-1 promotes growth and proliferation.Conclusion:tiRNA-1_33-Gly-GCC-1 is upregulated in colorectal adenocarcinoma tissue and may be a new oncogenic gene.Mechanically,tiRNA-1_33-Gly-GCC-1 can inhibit the expression of TP53I11 and promote the growth and proliferation of CAC cells.This study provides a new theoretical basis for the diagnosis,treatment,and prognosis of CAC in the future.
