Abstract:
Objective:To investigate the expression of autophagy related molecules mTOR,Beclin-1 and LC3 in benign and malignant pleural effusion,and to analyze their clinical significance.Method:Three hundred pleural effusion samples were included as research objects through liquid-based thin layer cytological examination and immunohistochemical examination combined with clinical data of patients,which were divided into group1(leaky pleural effusion) 65 cases,group2(non-tumor inflammatory pleural effusion) 142 cases,and group3 (non-small cell lung cancer pleural effusion) 93 cases.The gene and protein expressions of mTOR,Beclin-1 and LC3 in pleural effusion of each group were detected by RT-PCR and Western blot.Results:In terms of gene and protein expression,Beclin-1 and LC3 expression levels in group1 and group2 were increased compared with group3,while the expression of mTOR was significantly decreased,with statistical significance.At the same time,Beclin-1 and LC3 expression were decreased in group2 compared with group1,while mTOR expression was increased,the differences were statistically significant.In addition,the correlation analysis of benign and malignant pleural effusion showed that the relative expression value of Beclin-1 was positively correlated with LC3,while the relative expression value of mTOR was negatively correlated with Beclin-1 and LC3.Conclusion:There are differences in the expression of autophagy related molecules mTOR,Beclin-1 and LC3 in different types of pleural effusions,which may be related to the pathogenesis of inflammatory and non-small cell lung cancer caused by neoplastic pleural effusions,and can provide a new molecular basis for the identification of pleural effusions.
