Abstract:
Objective:The three cellular feature training sets were constructed based on HE-stained images of esophagogastric tumor tissue sections to auto-qualified the tumor-infiltrating lymphocytes(TILs) proportion for assisting in prognostic studies.Methods:The 214 adenocarcinoma of the esophagogastric junction(AEGJ),256 gastric adenocarcinoma(GAC),and 752 esophageal squamous cell carcinoma(ESCC) cases were included from Jiangsu Taixing.Each training set containing characteristics of tumor and stromal cells and lymphocytes was established by pathologists,and the verification of accuracy in cell recognition was performed using 2-fold validation,the correlation between single-cell annotations and automated recognition,and the comparison of auto-qualified TILs proportion with manual infiltration grade of TILs evaluated by pathologists.The Kaplan-Meier method and Cox proportional hazards model were conducted for prognostic value.Results:The image processing tools achieved a classification accuracy of 90% at least in TILs of esophagogastric tumors.The overall correlation coefficients between automated recognition and the pathologists’ quantitative assessment of AEGJ,GAC,and ESCC were 0.92,0.93,and 0.93,respectively,and the TILs correlation coefficients were 0.94,0.95,and 0.95,respectively.The automated recognition of the TILs proportion in AEGJ,GAC,and ESCC was consistent with the manual grading.Prognostic applications demonstrated the TILs proportion was an independent prognostic marker for esophagogastric cancers.Conclusion:This TILs infiltration assessment tool based on cellular training sets of esophagogastric cancers has a good application prospect in TILs assessment with prognostic research.
