Abstract:
Objective:To investigate the expression level of recombinant solute carrier family 7,member 11(SLC7A11) and microRNA-375(miRNA-375) in endometrial carcinoma tissues,and explore their correlation and clinical significance.Methods:40 cases endometrial carcinoma tissue and 30 cases endometrial tissue in normal proliferative membrance from January 2017 to January 2019 in Xuzhou Central Hospital were selected as research objects.Real-time fluorescence quantitative PCR(qRT-PCR) was used to detect the expression level of miRNA-375 and SLC7A11 mRNA.The expression level of SLC7A11 protein was detected by immunohistochemical.The correlation of miRNA-375 and SLC7A11 in endometrial carcinoma tissues was analyzed.The relationship between the two with the clinicopathological characteristics and the prognosis of patients was analyzed.Results:The expression level of miRNA-375 in endometrial carcinoma tissues was significantly lower than that in normal endometrial tissues(P<0.05).The expression level of SLC7A11 mRNA and positive expression rate of SLC7A11 in endometrial carcinoma tissues were significantly higher than those in normal endometrial tissues(P<0.05).The decrease of miRNA-375 expression level in endometrial carcinoma tissues was associated with the increase of FIGO stage,lymph node metastasis,muscle layer invasion and the increase of HE4,NLR and SII(P<0.05).The increase of SLC7A11 protein expression was associated with the increase of BMI,FIGO stage,muscle layer infiltration,CA125 and HE4(P<0.05).There was a negative correlation between miRNA-375 and SLC7A11 protein,SLC7A11 mRNA expression levels in endometrial carcinoma tissues(P<0.05).The 5-year cumulative survival rate of patients with low expression of miRNA-375 was lower than that of patients with high expression of miRNA-375(P<0.05),and the 5-year cumulative survival rate of patients with positive expression of SLC7A11 was lower than that of patients with negative expression of SLC7A11(P<0.05).The area under the curve of miRNA-375 and SLC7A11 mRNA expression in endometrial carcinoma tissues for the diagnosis of adverse prognosis were 0.764,0.859 and 0.875,respectively.The sensitivity was 0.767,0.700 and 0.833,respectively.The specificity was 0.775,0.950 and 0.900,respectively.The sensitivity of single index miRNA-375 was the highest,and the specificity of SLC7A11 mRNA was the highest.The specificity and sensitivity of the combination were high.Conclusion:The down-regulated expression of miRNA-375 and up-regulated expression of SLC7A11 in endometrial carcinoma tissues are often associated with poor prognosis of patients,and their combination may be have certain value in the diagnosis,treatment and prognosis evaluation of endometrial cancer.
