Abstract:
Objective:To investigate the expression of integrin α5 (ITGA5) in glioma tissues and its relationship with clinicopathological parameters and prognosis.Methods:Comparing the differences in the expression of ITGA5 and the biological information of clinical prognosis in glioma and normal brain tissues in the TCGA database.Collecting the glioma tissue specimens and clinical data of 54 patients with glioma surgery in Linyi People's Hospital,and the brain tissue specimens of 20 patients with traumatic brain injury after intracranial decompression.The brain tissue after trauma was used as the control,and the expression of ITGA5 protein in glioma was detected by immunohistochemical staining and western blotting.Statistical methods were used to study the correlation between ITGA5 expression level and clinicopathological parameters.By drawing survival curves,the difference in prognosis of patients with high and low expression of ITGA5 was compared.Results:In the TCGA database,the expression of ITGA5 in LGG and GBM was significantly different (P<0.001),and the expression increased with the increase of the WHO pathological grade of glioma,and was related to the poor prognosis of patients.The high expression rate of ITGA5 in glioma tissue was 59.2%(32/54),and the high expression rate in brain tissue after trauma was 5.0%(1/20),and the difference was statistically significant (P<0.05).The expression of ITGA5 in glioma tissue was correlated with WHO grade,Ki-67 and p53 genotype (P<0.05),but not with age,gender and tumor site (P>0.05).The survival time of patients with gliomas with high ITGA5 expression was significantly lower than that of patients with gliomas with low ITGA5 expression (P<0.05).Conclusion:The expression of ITGA5 is increased in glioma tissue,and the expression of ITGA5 is closely related to the pathological grade of glioma and the survival of patients.
