Abstract:
Objective:To investigate the hsa-miR-769(miR-769) expression in esophageal squamous cell carcinoma and its effect on the proliferation,invasion,migration and paclitaxel resistance of cancer cells.Methods:Through bioinformatics technology mining,we conducted an in-depth study on the expression of tumor-associated hsa-miR-769(miR-769),and further verified clinical samples,and further explored its mechanism of action through cell biology and molecular biology studies.Results:The mining results of public database showed that miR-769 expression was abnormally elevated in esophageal carcinoma tissues,and the same abnormal expression was observed in clinical samples.Through cell experiments and molecular biology experiments,we found that miR-769 regulated the proliferation of tumor cells invasion and migration ability can also cause changes in the epithelial mesenchymal transformation process of tumor cells.In subsequent studies,we successfully constructed a paclitaxel-resistant esophageal squamous cell carcinoma line,and found that miR-769 played an important role in the acquired resistance of esophageal squamous cell carcinoma to paclitaxel.Conclusion:miR-769 affect the proliferation,invasion,migration and resistance to paclitaxel of esophageal squamous cell carcinoma.This study aims to explore the mechanism of the occurrence and development of esophageal squamous cell carcinoma,and provide a new potential target for targeted therapy of esophageal squamous cell carcinoma.
