Abstract:
Objective:To explore the effects of miR-409-3p and its target gene on autophagy,growth and metastasis of small cell lung cancer cells.Methods:qRT-PCR was used to detect the expression of miR-409-3p in HPAEpiC,SBC-5,NCI-H446,HOP-92 cells.miR-409-3p mimic and miR-NC were transfected into SBC-5 cells,and the CCK8 method was used to detect cell proliferation.Transwell chamber method was used to detect cell migration and invasion.Flow cytometry was used to detect cell apoptosis.Dual luciferase reporter gene experiment was used to detect the targeted binding of miR-409-3p and Beclin-1.After transfecting with miR-409-3p mimic,the Beclin-1 overexpression plasmid (miR-409-3p mimic+Beclin-1) was transfected to SBC-5 cells,and then the levels of cell proliferation,migration,invasion and apoptosis of each group were detected.Western blot was used to detect the expression of Beclin-1,LC3-Ⅱ,and p62 in each group of cells.Results:The expression of miR-409-3p in SBC-5,NCI-H446,and HOP-92 cells was significantly lower than that in HPAEpiC cells.The cell proliferation,migration and invasion ability in miR-409-3p mimic group was significantly lower than that in miR-NC group,and the level of apoptosis was significantly higher than that in miR-NC group.The test results of dual luciferase reporter gene experiment showed that miR-409-3p targeted Beclin-1.The cell proliferation,migration and invasion ability of miR-409-3p mimic+Beclin-1 group was significantly higher than that in miR-409-3p mimic group,and the level of cell apoptosis was significantly lower than that in miR-409-3p mimic group.Beclin-1 expression and LC3-Ⅱ expression in miR-409-3p mimic group were significantly lower than miR-NC group,and p62 expression was significantly higher than miR-NC group.The LC3-Ⅱ expression of the miR-409-3p mimic+Beclin-1 group was significantly higher than that in the miR-409-3p mimic group,and the p62 expression was significantly lower than that in the miR-409-3p mimic group.Conclusion:miR-409-3p regulates the target gene Beclin-1 to inhibit the growth and metastasis of SBC-5 cells.The mechanism may be the inhibition of autophagy in small cell lung cancer.
