Abstract:
Objective:To explore the expression and prognostic significance of lncRNA ALMS1-IT1 in colorectal cancer (CRC) and determine the impact of ALMS1-IT1 in CRC cell proliferation,invasion and epithelial-mesenchymal transition (EMT).Methods:The expression of ALMS1-IT1 in CRC and its correlation with the survival of patients were analyzed by GEPIA web server using The Cancer Genome Atlas (TCGA) database.The expression of ALMS1-IT was detected by RT-qPCR in human normal intestinal epithelial cells (NCM460) and CRC cells (HCT116,HT29,LoVo,SW480).ALMS1-IT was silenced and overexpressed by plasmid transfection in LoVo and SW480 cells,respectively.CCK-8 assay detected cell proliferation ability.Transwell assay analyzed cell migration and invasion potentials.Immunoblotting was used to detect the changes of EMT markers (E-cadherin,N-cadherin,Vimentin).Results:TCGA data analysis confirmed that ALMS1-IT expression in CRC tissues was significantly higher than that in normal tissues,and high expression of ALMS1-IT1 was prominently associated with poor prognosis.The expression of ALMS1-IT1 in CRC cells (HCT116,HT29,LoVo,SW480) was markedly higher than that in normal intestinal epithelial cells (NCM460).ALMS1-IT1 silencing inhibited cell proliferation,migration,invasion,and led to increased expression of E-cadherin protein and decreased expression of N-cadherin and Vimentin protein in LoVo cells.Overexpression of ALMS1-IT1 promoted cell proliferation,migration,invasion,and resulted in decreased expression of E-cadherin protein and increased expression of N-cadherin and Vimentin protein in SW480 cells.Conclusion:ALMS1-IT1 is highly expressed in CRC tissues and is closely related to poor prognosis.ALMS1-IT1 exerts an oncogenic role by promoting the proliferation,invasion and EMT of CRC cells.
