Abstract:
Objective:To investigate the expression of Golgi phosphoprotein 2(GOLM1) in lung adenocarcinoma tissues and the effect on proliferation,migration and invasion of A549 cells.Methods:The expression of GOLM1 mRNA in lung adenocarcinoma tissues and its correlation with prognosis were analyzed from GEPIA database.Lentiviral siRNA technology was used to down regulate the expression of GOLM1 in human lung adenocarcinoma A549 cells.Real-time PCR was used to verify the silencing effect.CCK-8,cell scratch test and Transwell test were used to detect the cell proliferation,migration and invasion.Results:GOLM1 mRNA was highly expressed in lung adenocarcinoma(P<0.05).The overall survival time of patients with high GOLM1 mRNA expression was significantly lower than that of patients with low GOLM1 expression(P<0.05).The proliferation,migration and invasion of A549 cells were significantly decreased after GOLM1 knockdown(P<0.05).Conclusion:GOLM1 is highly expressed in lung adenocarcinoma,silencing its expression can inhibit cell proliferation,migration and invasion of lung adenocarcinoma cells.It can be used as a potential prognostic marker and therapeutic target for lung adenocarcinoma.
