Abstract:
Objective:To investigate the mechanism of the cytotoxicity of NK and NKT cells enhanced by DC from umbilical cord blood on leukemia cells.Methods:Umbilical cord blood mononuclear cells (PBMC) were extracted and cultured into DC,NK and NKT lymphocytes in vitro.The killing effects of NK,NKT,NK+DC and NKT+DC on target cells were detected by using human myeloid leukemia K-562 cells as target cells.The level of IFN-γ in the co-culture supernatant was detected.Results:The killing effect of NKT+DC on target cells was significantly enhanced compared with that of NKT group (P<0.05).The killing effect of NK+DC on target cells was also significantly stronger than that of the NK group (P<0.05).The expression level of IFN-γ was the highest in the NKT+DC+target cell group compared with other groups,and reached the highest at 72 h (P<0.05).The expression of IFN-γ in the NK+DC+target group was higher than that in the NK+target group (P<0.05).Conclusion:DC can effectively enhance the killing effect of NKT cells and NK cells on leukemia cells by stimulating NKT cells to produce a large amount of IFN-γ.
