Abstract:
Objective:To investigate the prognostic significance of angiopoietin 1(ANGPT1) and the exprssion of ANGPT1 in lung adenocarcinoma tissues.Methods:Data of lung adenocarcinoma and lung squamous cell carcinoma with the ANGPT1 gene expression profile data and clinical information were downloaded from the Cancer Genome Atlas(TCGA) database.The difference of ANGPT1 expression in lung cancer tissues and normal tissues was determined by t test.The correlation between ANGPT1 gene expression and clinicopathological features of lung adenocarcinoma patients was analyzed by χ2 test.Log-rank test was used for survival analysis.To predict the pathways that may be regulated by ANGPT1 in lung adenocarcinoma,Gene Set Enrichment Analysis(GSEA) method was used for gene set enrichment analysis.Results:The mRNA level of ANGPT1 in lung cancer tissues was lower than that in normal tissues.Its expression level was negatively correlated with T stage(P=0.013).Patients with low-expression ANGPT1 lung adenocarcinoma had significantly worse prognosis than those with high expression(P=0.006).ANGPT1 was an independent protective factors for adenocarcinoma prognosis(P<0.05).GSEA results showed that ANGPT1 might affect the development of lung adenocarcinoma by regulating G2M checkpoints,DNA damage repair,apoptosis,and cell proliferation-related pathways such as mTOR signaling,MYC signaling and mitotic spindle.Conclusion:ANGPT1 is an independent prognostic factor for lung adenocarcinoma and might be as a potential prognostic biomarker and therapeutic target for lung adenocarcinoma.
