Abstract:
Objective:To investigate the effect of pemetrexed and sunitinib on the proliferation,cell cycle and apoptosis of hepatoma cells.Methods:After pemetrexed and sunitinib single or combined treatment of hepatocellular carcinoma SK-Hep1 cells,the inhibition rate of cell proliferation was detected by MTT assay,and cell cycle and apoptosis rate were tested by FCM,and the proteins of kinase B (AKT),phosphorylated protein kinase B (p-AKT) and proliferating cell nuclear antigen (PCNA),B cell lymphoma -2 gene (Bcl-2) and cutting type caspase 3 protein expression (Cleaved caspase-3) were measured by Western blot.Results:Pemetrexed and sunitinib were able to inhibit the proliferation of SK-Hep1 cells in a time-dependent manner,and the IC50 in 72 h time was (2.89±0.20)μmol/L and (2.12±0.12)μmol/L,respectively.Pemetrexed and sunitinib could both induce apoptosis in SK-Hep1 cells.Pemetrexed blocked the SK-Hep1 cell cycle in S phase,and sunitinib blocked the cell cycle in G0/G1 phase.The expressions of p-AKT,PCNA and Bcl-2 were decreased and the expression of Cleaved caspase-3 was increased by pemetrexed and sunitinib.The combination of two drugs made SK-Hep1 cells arrest in G2/M phase.The proliferation inhibition rate and apoptosis rate of cells were more obvious than that of single drug,and the expression of p-AKT,PCNA and Bcl-2 decreased,and the expression of Cleaved caspase-3 increased more significantly.Conclusion:Pemetrexed combined with sunitinib can inhibit the proliferation of SK-Hep1 cells and induce SK-Hep1 cell cycle arrest and apoptosis.The mechanism may be related to the down-regulation of p-AKT,PCNA,Bcl-2 expression and upregulation of Cleaved caspase-3 expression.
