Abstract:
Objective:To study the effect of silencing MTA1 on the adhesion,migration and invasion on hepatocellular carcinoma cells.Methods:MTA1 siRNA and siRNA control were transfected into hepatoma cells,at the same time,the cells without transfection were used as control.qRT-PCR and Western blot were used to detect the expression level of MTA1.MTT detection of cell proliferation,cell adhesion test was used to detect cell adhesion.Transwell was used to detect cell invasiveness and migration.Western blot was used to detect the levels of MMP-2,MMP-9,E-cadherin and N-cadherin protein in cells.Gelatinase zymography was used to detect the activity of MMP-2 and MMP-9.Results:Transfection of MTA1 siRNA in cells could downregulate the level of MTA1 mRNA and protein in hepatoma cells.Transfection of siRNA control had no effect on MTA1 mRNA and protein level in cells.After silencing MTA1,the survival rate and adhesion rate of hepatoma cells were decreased.The number of invasive cells was reduced.Compared with cells without transfection,the difference was statistically significant (P<0.05).After silencing MTA1,the levels of MMP-2,MMP-9 protein and activity were decreased,at the same time,the level of E-cadherin protein was increased,and the level of N-cadherin protein was reduced.Compared with cells without transfection,the difference was statistically significant (P<0.05).Conclusion:Silencing MTA1 inhibits the adhesion,migration and invasion of hepatocellular carcinoma cells.The mechanism is related to the inhibition of cell secretion of MMP-2,MMP-9 and inhibition of EMT.
