Abstract:
Objective:To investigate the inhibition of E- cadherin(E-cad) expression by Snail protein in invasion and lymph node metastasis of epithelial ovarian cancer cells.Methods:Human ovarian cancer cell lines C13 were selected,the Snail/siRNA and Snail/smRNA were transfected into C13 cells,used the PBS as the blank control group,the expression of E-cad protein were detected,cell invasion and lymph node metastasis were given experimental analysis by Transwell chamber.Results:The 12 h transmembrane C13 cell numbers in the blank control group,Snail/smRNA group,Snail/siRNA group were 98.24±7.92,90.93±8.38 and 21.48±4.85,there were significant difference in Snail/siRNA group compared with the other two groups(P<0.05),the cell transfer in the Snail/siRNA group compared the other two groups were also significant difference(P<0.05).Compared with the Snail/smRNA group,the protein levels of E-cad in Snail/siRNA group was decreased,and the difference compared between the two groups was statistically significant(P<0.05),while the blank control group did not change significantly(P>0.05).Conclusion:Inhibition of E- cadherin expression by Snail protein can inhibit the invasion and lymph node metastasis of epithelial ovarian cancer cells,it can provide theoretical evidence for the targeted treatment and reference target for the epithelial ovarian cancer.
