Abstract:
Objective:To investigate the effects of arsenic trioxide (As2O3) of various concentrations on invasion,migration and USP22 expression in human HepG2 hepatocarcinoma cells and the underlying molecular mechanism.Methods:CCK8 was used to test the proliferation levels of HepG2 cells treated with As2O3 of different concentrations in different time.Wound-healing and Transwell assays were used to examine the effects of As2O3 of different concentrations on cell migration and invasion.The protein levels of USP22 treated with As2O3 of different concentrations were determined by Western blot.Results:With the increase of As2O3 concentration and time,the proliferation of HepG2 cells was obviously inhibited.The migration,invasion and the expression of USP22 protein in HepG2 cells which were treated with 2,4,8 μmol/L As2O3 respectively for 24 h were obviously decreased compared with the control group.Conclusion:As2O3 could inhibit the migration and invasion of human hepatocellular carcinoma HepG2 cells,which may be related to the down-regulation of USP22 protein.
