Abstract:
Objective:To isolate the upstream transcriptional regulatory region of MLAA-22 gene,identify promoter activities of the isolated regions.Methods:-999~+1 and-1999~+1 of MLAA gene' s upstream transcriptional regulatory region were synthesized and inserted into BglⅡ and NheI restriction sites of dual-Luciferase reporter gene vector psiCHECK to construct recombinant plasmids.The recombinant plasmids were named psiCHECK-2-MLAA-22(-999~+1) and psiCHECK-2-MLAA-22(-1999~+1) respectively.The promoter activities of isolated regions were detected by dual-Luciferase reporter gene system.Results:Both MLAA-22(-999~+1) and MLAA-22(-1999~+1) displayed high luciferase activity.Besides,the luciferase activity of MLAA-22(-999~+1) of was stronger than that of MLAA-22(-1999~+1).Conclusion:The (-999~+1) region will be the key region for the study of promoter activity of MLAA-22 gene' s upstream transcriptional regulatory region in future.
