Abstract:
Objective:To investigate the effect and mechanism of ghrelin regulated ERK signaling pathway on multidrug resistance of breast cancer cells.Methods:Human breast cancer cell MDA-MB-231 cell culture,to set up control group,doxorubicin group,ghrelin group,ghrelin combined with doxorubicin group,ghrelin combined with doxorubicin plus PD098059 inhibitor group,flow cytometry was used to detect cell apoptosis.The expression of ERK,p-ERK and P-gp protein was detected by Western-blot method.Results:Human breast cancer MDA-MB-231 cell apoptosis rate was lowest by ghrelin intervention(P<0.01),and highest by doxorubicin intervention (P<0.01).Ghrelin combined with doxorubicin could inhibit the pro apoptotic effect of doxorubicin on human breast cancer MDA-MB-231 cells (P<0.01).Ghrelin combined with doxorubicin plus ERK signaling pathway specific blocker PD98059 could attenuate the inhibitory effect of ghrelin combined with doxorubicin on human breast cancer MDA-MB-231 cell apoptosis(P<0.01).Compared with ghrelin group,the expression of ERK and the level of p-ERK were significantly decreased in doxorubicin group(P<0.05),and P-gp protein expression of ghrelin group compared with the doxorubicin group was significantly increased(P<0.05).The expression of ERK and the level of p-ERK of ghrelin combined with doxorubicin group compared with doxorubicin group were significantly increased(P<0.05),and the expression of P-gp protein was significantly increased(P<0.05).The expression of ERK and the level of p-ERK of ghrelin combined with doxorubicin plus inhibitor group compared with ghrelin combined with doxorubicin group were significantly decreased(P<0.01),and there was no significant difference in the expression of P-gp protein between the two groups(P=0.07).Conclusion:Certain concentration of ghrelin activates ERK signaling pathway in breast cancer cells to increase P-gp protein expression,inhibit apoptosis of breast cancer cells by doxorubicin and induce drug resistance.Blocking the ghrelin-ERK signaling pathway may reverse the occurrence of multidrug resistance in breast cancer cells.
