Abstract:
Objective:To observe the inhibitory effect of Apigenin on the Hela cell lines,and investigate the potential mechanism.Methods:MTT assay was used to detect the anti-proliferative effect of Apigenin on Hela cells.The influence of Apigenin on growth curve of Hela cells was detected by real time cellular analysis.The PI3K/Akt signal pathway was analyzed by Western blot assay.Results:Compare with negative control,apigenin showed a significantly inhibitory ability to Hela cells (P<0.01).After treated with Apigenin for 48h,the IC50 value to Hela cells were (6.8±0.8)μmol/L,inhibition rate rised to (80.5±5.3)% while the concentration is 20μmol/L.Cell growth curve indicate that Apigenin is able to slow down the proliferation of Hela,after treated with 20μmol/L Apigenin,the cell count were quickly reduced.Apigenin could intensively resist Akt activation which caused by IGF-1,but had no effect on PI3K,Apigenin could also launch the mitochondrial apoptotic by reducing the Bcl-2/Bax proportion and increasing the activity of caspase family.Conclusion:Apigenin could inhibit the proliferation of Hela cells and induce apoptosis,the potential mechanism is probably related with regulation of PI3K/Akt pathway.
