Abstract:
Objective:To study the effect of triptolide(TPL) on expression of ID4 and DNA methyltransferases(DNMT) mRNA on lymphoma Raji cells.Methods:Afer treated with 0,20ng/ml TPL for 24h,the Raji cells' mRNA expression of ID4,DNMT1,DNMT3A,DNMT3B was detectected by Fluorescent quantitative RT-PCR.Afer treated with 0,5,10,15,20ng/ml TPL for 24h,the Raji cells' mRNA expression of DNMT1,DNMT3A,DNMT3B was detectected by Fluorescent quantitative RT-PCR.Results:ID4 gene failed to express in Raji cells without TPL.The expression was recovered after the cells exposed to 20ng/ml TPL.TPL could significantly down-regulate DNMT1,DNMT3Aand DNMT3B gene on mRNA level in a dose dependent manner.Conclusion:TPL can inhibit the Raji cell DNA methyl transferase,activate the expression of ID4 mRNA.
