The regulation of Wnt3a on the cell growth in non-small-cell lung cancer cell lines

Objective:To approach the molecular mechanism of Wnt signaling pathway in non-small-cell lung cancer(NSCLC),and the effect of recombinant human Wnt3a on NSCLC cell lines.Methods:H460 and A549 cell lines were treated with recombinant human Wnt3a.The alteration of the cell numbers,colony forming in soft-agar and cell cycle were observed.Results:The treatment of recombinant human Wnt3a significantly increased cell numbers [(4.26±0.26)×105/ml vs (2.42±0.44)×105/ml in A549,P<0.01;(3.98±0.21)×105/ml vs (2.07±0.10)×105/ml in H460,P<0.01]and the efficiency of colony forming[(1.57±0.11)% vs (1.03±0.08)% in A549,P<0.01;(1.53±0.11)% vs (0.79±0.10)% in H460,P<0.01].The flow cytometric analysis showed that the cells of G0/G1 phase declined and the cells of S phase rose after the treatment of recombinant human Wnt3a.Conclusion:As an activator of canonical Wnt signaling,recombinant human Wnt3a may promote cell growth through its regulation to cell cycle in NSCLC cell lines.
