Abstract:
Objective:To explore the application value of three novel serum markers,COL10A1,GKN2,and LIPF,in diagnosing gastric cancer (GC).Methods:The differential expression biomarkers of GC were mined as candidate markers through the public databases (TCGA and GTEx).A total of 108 individuals who underwent gastroscopy and pathological examination in our hospital from November 2020 to October 2022 were enrolled,including 48 healthy individuals and 60 patients with GC [28 cases in the early GC stage (EGC group) and 32 cases in the advanced stage (AGC group)].The correlation between serum levels of markers and the incidence of GC was analyzed,and receiver operating curve (ROC) was drawn,and the application value of single and combined detection of novel markers and conventional markers (PGI,PGII) detection in the diagnosis of GC was analyzed.Results:Three GC differential expression proteins (COL10A1,GKN2,and LIPF) were mined through public databases.The results of testing three markers in blood samples showed that the area under the curve (AUC) of the three-marker combined panel (COL10A1+GKN2+LIPF,3MP) in distinguishing GC (3MP vs PGR:0.842 vs 0.841) and EGC (3MP vs PGR:0.877 vs 0.843) from high-risk populations were all higher than PGR (PGI/PGII).The AUC of combined detection of five markers (3MP+PGI+PGII,5MP) in detecting GC and EGC was 0.963 and 0.953,respectively.Conclusion:COL10A1,GKN2 and LIPF are potential serum markers for the diagnosis of GC.The combined detection of the three novel markers and PGI,PGII has important reference value for the early detection of GC.
