Abstract:
Objective:To investigate the differential expression of ILK protein in cancer tissue between BRAF mutant and non mutant thyroid cancer patients,and to explore the clinical significance of ILK protein.Methods:Basic information of 46 patients diagnosed with papillary thyroid carcinoma after thyroidectomy in our hospital from December 2020 to June 2023 was collected.Differences in BRAF gene mutations and pathological parameters such as gender,age,primary lesion diameter,tumor distribution,TI-RADS grading by ultrasound,TNM and pathological staging of cancer,invasion of the thyroid capsule and lymph nodes by the primary lesion,and presence of Hashimoto's thyroiditis were statistically analyzed for all enrolled patients.Immunohistochemical staining was performed on the primary cancer slices of all enrolled patients to evaluate the strength of ILK protein.Based on the previous detection results,the grading of ILK protein strength in 46 tissues was recorded,and the correlation between ILK and various clinical pathological parameters of PTC was analyzed.Statistically analyze the relationship between the intensity of ILK expression and whether PTC patients have BRAF V600E gene mutations.Results:BRAF V600E gene mutation was positively correlated with PTC multiple cancer lesions,higher stage and grade,lymph node and extracapsular infiltration (P<0.05),but not with gender,age,TI-RADS grade,primary lesion diameter,or presence of Hashimoto's thyroiditis (P>0.05).ILK protein shows varying degrees of positive expression in epithelial papillary tissues,and strong positive signals are closely related to malignant biological behaviors such as larger tumor diameter,multifocal infiltration,lymph node metastasis,and extracapsular invasion (P<0.05).The strong positive rate of ILK protein detected in the primary cancer lesions of BRAF mutant PTC patients is much higher than that of non mutant patients (P<0.05).Conclusion:ILK protein is highly expressed in BRAF mutant thyroid cancer and is closely related to malignant behaviors such as larger tumor diameter,multifocal invasion,lymph node metastasis,and extracapsular invasion.It is involved in the process of cancer invasion induced by BRAFgene mutation.
