Abstract:
Objective:To investigate the expression of human leukocyte antigen-DRB1 (HLA-DRB1) in hepatocellular carcinoma (HCC) tissues and its clinical significance.Methods:HLA-DRB1 protein expression level in HCC tissues and adjacent tissues from 343 patients was assessed using immunohistochemistry,and clinicopathological characteristics of patients with different HLA-DRB1 expression levels were compared.Kaplan-Meier method was used to compare the overall survival (OS) and recurrence-free survival (RFS) of patients with different HLA-DRB1 expression levels.Cox proportional hazards regression was employed to analyze the factors influencing overall survival (OS) and recurrence-free survival (RFS) in HCC patients after hepatectomy.Results:The high expression rate of HLA-DRB1 protein in HCC tissues (47.23%) was significantly lower than that in adjacent tissues (79.59%,χ2=77.415,P<0.001).HLA-DRB1 expression level was associated with AFP>400 μg/L,cirrhosis,no/incomplete capsule,microvascular invasion,and BCLC B/C stage (all P<0.05).The OS rate (χ2=4.486,P=0.034) and RFS rate (χ2=15.446,P<0.001) of HCC patients with high expression of HLA-DRB1 were significantly higher than those with low expression of HLA-DRB1.The high expression of HLA-DRB1 protein was an independent protective factors affecting postoperative OS (HR=0.727,95%CI:0.531~0.944,P=0.040)and RFS(HR=0.607,95%CI:0.454~0.811,P<0.001) in HCC patients.Conclusion:The high expression rate of HLA-DRB1 protein in HCC tissues was notably lower than that in adjacent tissues,and high expression of HLA-DRB1 protein was significantly associated with good prognosis of HCC patients.
