Abstract:
Objective:To investigate the expression of microRNA (miR)-574-3p and family with sequence similarity 3 member C (FAM3C) in esophageal cancer tissue and their relationship with clinicopathological characteristics in esophageal cancer patients.Methods:From May 2018 to May 2020,90 patients with esophageal cancer who underwent surgical treatment in our hospital were collected,and specimens of cancer tissue and adjacent tissue were collected during the surgery.The qRT-PCR method was applied to detect the relative expression levels of miR-574-3p and FAM3C mRNA in tissues.Immunohistochemical method was applied to detect the expression of FAM3C protein in tissues.Spearman rank correlation was applied to analyze the correlation.The relationship between the expression levels of miR-574-3p and FAM3C proteins and the clinical pathological characteristics of patients was analyzed.Kaplan-Meier method was applied to analyze the relationship between miR-574-3p and FAM3C protein expression levels and prognosis.Results:Low expression of miR-574-3p and high expression of FAM3C mRNA and FAM3C protein in cancer tissues of patients with esophageal cancer (P<0.05).There was a negative correlation between miR-574-3p and FAM3C(r=-0.420,P<0.05).Pearson correlation analysis showed that the mRNA expressions of miR-574-3p and FAM3C in esophageal cancer tissues were negatively correlated (r=-0.731,P<0.05).miR-574-3p and FAM3C proteins were correlated with TNM stage,invasion degree and lymph node metastasis (P<0.05).The 3-year cumulative survival rate of the miR-574-3p high expression group was 83.72%,which was obviously higher than that of the miR-574-3p low expression group (53.19%).The 3-year cumulative survival rate of the FAM3C high expression group was 56.86%,obviously lower than that of the FAM3C low expression group (82.05%) (log rank χ2=10.175,7.290,P<0.05).Conclusion:The expression level of miR-574-3p is low in esophageal cancer tissue,while the expression level of FAM3C is high,and there is a negative correlation between the them,which is related to clinical pathological features such as TNM staging,invasion degree,lymph node metastasis,and 3-year cumulative survival rate of patients.
