Abstract:
Objective:To screen the differentially expressed LncRNA in plasma extracellular vesicles (EVs) of breast cancer (BC),and to verify its diagnostic value for BC.Methods:Through electron microscopy,particle size analysis and Western blot,EVs in plasma samples were identified.The plasma EVs of 8 BC patients and 8 healthy people were analyzed by LncRNA sequencing.The expression of LncRNA Linc01305 in plasma EVs of 65 BC patients,65 healthy control samples,and patients with differential diagnosis of breast fibroadenoma,cystic hyperplasia,and plasma cell mastitis were detected.The expression of Linc01305 in breast cancer cell line was detected by qRT-PCR.The expression level of Linc01305 in plasma EVs of BC patients before and after surgery was detected by qRT-PCR.The correlation between the expression of Linc01305 in BC patients and clinical data was statistically analyzed.Results:Electron microscopy,particle size analysis and western blot detection identified typical characteristics of exosomes.Sequencing results showed that the expression of LncRNA Linc01305 in plasma EVs from BC patients was higher than that of healthy controls.The sequencing results were confirmed by qRT-PCR.Linc01305 was highly expressed in BC cancer cell lines (P<0.01),but decreased in BC patients after tumor resection (P<0.01).The high expression of was correlated with tumor size,infiltrative staging,lymphatic metastasis and TNM stage.The area under the curve (AUC) of Linc01305 in the diagnosis of BC patients was 0.771 2(P<0.000 1).Conclusion:LncRNA Linc01305 was highly expressed in plasma EVs of BC patients and also related to tumor size,infiltrative staging,lymphatic metastasis and TNM stage,which is expected to become a new diagnostic marker for BC.
